美国南卡罗莱纳医科大学成立于1824年，至今有近200年的历史，从一家小型私人医院逐渐发展成为目前美国高级学术医学中心之一，当时是一所致力于为内科医生提供培训的小型私立学院。这是美国第六古老的目前仍在运营的医学院，也是南方地区最古老的一所。目前南卡罗莱纳医科大学已经扩大到一所州立学校，拥有一个医学研究中心和6个专门为健康专业人士、生物医学科学家和其他与健康有关的人员提供广泛教育服务的学院。学校同时经营者一所研究所和一个公立医院。
